They encounter new races, new threats, maybe even some internal strife between the various Citadel colonists. Many many years/centuries/whatever pass before they arrive at Andromeda (you could explain this away with time relativity, cryo-sleep, whatever) and they have no knowledge of how the events in the Milky Way eventually panned out. A group of colonists consisting of the Citadel races, maybe led by some Spectres, set off to travel from the Milky Way to Andromeda, either before the Reaper threat is discovered or afterwards as a contingency plan in case the Reapers are successful. I think it would be pretty cool if they treated this a bit like the Outbound Flight Project from Star Wars (only more successful). Strong opportunity for commentary around native rights and displacement! That said, the trailer is extremely wild west, so colonisation seems like the obvious theme. It lets us know to expect in terms of some of the rules that exist, the play style, and the visual and aural style.

Would eliminate any baggage that fans of the series might have left and open up their storytelling to new fans that never got into ME. If they don't tie into, in some way, the history of the world there's no reason for them to not create new justifications for galactic travel (or simply ignore it all together) and create a new universe to play in. You are right in that Shepard is merely the avatar of storytelling though and can be replaced. Everything from Saren's attack on Eden Prime to the final "choice" centers on the Reapers. Saying the series is about the technology is like saying Star Trek is about Warp Drive. The technological macguffins that allow the universe to exist was merely that. The series is about the impending onslaught of the Reapers and how the galaxy will survive them.
